    91128 PALAISEAU Cedex 2013 - 2017 This project aims to develop droplet-based microfluidic systems to incorporate nucleic acids into cationic liposomes and to transfect mammalian cells in vitro. The first step of the project took place in Brazil and involved the development of a microfluidic droplet system that promotes rapid mixing in order to incorporate pDNA into cationic liposomes, generating reproducible and suitable complexes for biological applications, like gene therapy. The second step of the project took place in France and involved the development of a microchip to transfect mammalian cells in vitro within soft 3D droplets of hydrogel, using the complexes as nanovectors. With this system, it is possible to investigate key parameters that influence the process of transfection in mammalian cells, which could be a potential tool for therapeutic vaccines.

    2010 - 2013 This research aimed at the development of a production process for cationic liposomes for use in cancer immunotherapy strategies. Dendritic cells (DCs) vaccine has the function of helping the immune system to identify tumor antigens in order to eliminate cancerous cells. Cationic liposomes, in addition to activated DCs, are able to safely mediate the introduction of nucleic acids into cells. Through this strategy, total RNA extracted from tumor cells were incorporated into cationic liposomes, which were introduced in vitro into DCs. As a result, we developed a scalable process to produce cationic liposomes with physico-chemical characteristics that lead to their internalization by DCs, stimulating and delivering nucleic acids to cells.
