Team '' Oxidative Stress, Redox Regulation and Cell Fate''
Mounting evidence has established that reactive oxygen species (ROS) homeostasis and dynamic intracellular reductive/oxidative (redox) balance play a key role in maintaining genome stability and regulating cell survival/death. Defects in ROS homeostasis and redox signaling are at the crux of pathological process such as aging, neurodegenerative diseases, cancer initiation, progression and drug resistance. In fact one of the main features of cancer cells, when compared to the normal ones, is a persistent pro-oxidative state that can lead to intrinsic oxidative stress. However, cancer cells have evolved mechanisms to protect themselves from this intrinsic oxidative stress and have developed a sophisticated adaptation system that essentially involves the rearrangement of the antioxidant functions and the upregulation of pro-survival molecules. Despite numerous efforts, our knowledge of the spatio-temporal pattern of intracellular redox changes that is highly non-uniform between and within cellular compartments remains preliminary. In this context, the goal of my internship was to reveal that DNA replication inhibitor hydroxyurea alters Fe-S centers by producing reactive oxygen species in vivo. In order to set up a therapeutic strategy based on the modulation of ROS levels/redox homeostasis in cancer cells.

Basal cell carcinoma (CBC) is most commonly due to genetic mutations caused by UV. Those mutations most often affect the TP53 tumor suppressor gene, this deficiency being responsible for 56% of the CBC.
However, our research shown that a large majority of the West Algerian population affected by CBC owns mutations in the patched gene (PTCH1).
The goal of my internship was the study of the involvement of the polymorphism Leu1315Pro, gene Ptch in the development of basal cell carcinoma in the Algerian population. In order to develop a new chemotherapy more adapted to the West Algerian patients affected by the CBC.
