LAVAUR

En résumé

As a research scientist with 10 years experience in R&D in the pharmaceutical industry and 2 years in a biotechnology, I manage projects with main expertise in development of animal and cellular models for the assessment of drug efficacy in different therapeutic areas (neurology, cardiovascular, oncology and dermatology).

The implementation of these models includes pharmacological validation, analysis of biomarkers to validate target activity and study of molecular drug signature. I also participate in the selection of new therapeutic targets.

These missions involve management of technicians and trainees, coordination and planning of studies, analyse and synthesis of results, report writing and communication.

Main skills: molecular and cellular biology (differential analysis of gene expression by q-RTPCR, reporter-gene assays, primary culture of hepatocytes, culture of different cell lines, target invalidation by siRNA), western-blotting, immunohistochemistry, in vivo experiments (pharmacological treatments, tissue sampling).

Entreprises

  • P. Fabre - Research scientist in molecular and cellular biology

2010 - maintenant Over the last three years, I have been responsible for the implement and supervision of a new model and the compound screening destined to a topical application on a new ex-vivo model. I had to develop and demonstrate the effectiveness of this model. I am also in charge of coordinating the different actors involved in this project, analyse and present data, and manage molecular and immunohistochemistry technique on skin biopsies.

Animal model development for assessment of drug efficacy in dermatologic field
    Pharmacological validation of the model
Skin sampling and molecular biomarkers analysis (qPCR and Immuno-Histo-Chemistry)
    Project Management
    Competitive intelligence
Involvement in drug-discovery and characterisation of therapeutic drugs (metabolism, neurology, oncology)
Biomarker search for validating target activity
Study of molecular signature of drugs / pathology
    Personal and technical management
    Oral communications / reporting
Participation in the selection of therapeutic target (bibliographic search)
  • Pierre Fabre - Research associate – PhD student

Castres 2006 - 2010 During my PhD position, I capitalized a lot of experiment on primary culture of rat hepatocytes to mimic the metabolic deregulations induced by antipsychotic drugs. The physiologic and pharmacological validations of this model demonstrated the strong power of primary culture versus other hepatic cell lines. Thanks to a rigorous scientific approach, we demonstrated a potential mechanism of action involved in these metabolic deregulations. This work, in collaboration with Inserm UMR 1011 (Bart Staels, Lille, France) and Institute of Predictive Medicine and of Therapeutics Research-Faculty of Medicine (Patrick Duriez, Lille, France), resulted in 4 publications included 3 international journals.
  • Pierre Fabre - Molecular biology Engineer

Castres 2004 - 2006 I developed an ex-vivo screening test for a cardiometabolic program. I trained and managed one technician on RNA tissue extraction, RT and qPCR techniques. I also worked for the Neuropsychiatric axis to implement a platform for differential expression analysis of 40 receptors by qPCR on brain tissues. This challenge required a vast bibliographic study and an essential period to design and validate primers. Moreover, I developed cell models by transfecting different nucleic acids (siRNA, plasmids) to study the impact of G subunits on the GPCR signal transduction.
  • Proteus - Engineer in molecular biology

2004 - 2004 Development of automated nucleic acid purification technical for quantitative PCR amplification of bacterial RNA from air/liquid/solid specimens (Genesis Tecan, RotorGene from Corbett Research)
  • Biomerieux (Boston, USA) - MA training

2001 - 2001 Participation in the development of « VIDAS Probe » technical for the molecular diagnostic of N. gonorrhoeae from biological specimens. Oral presentation and report redaction (English)
  • Exonhit therapeutics - Molecular Biologist

2001 - 2003 2 year-experience work in a biotechnology company (ExonHit Therapeutics) in Paris, France in the oncology field : “Identification of splicing isoforms regulated by the VHL oncogene in a cancer renal cell line and in biopsies of human renal carcinoma”.
We managed to identify some biomarkers (splicing isoforms) regulated by the VHL oncogene and differentially expressed in renal carcinoma compared to normal tissues.
RNA and DNA extraction, RNA differential expression analysis (macro-array, qPCR, Northern Blot), cell culture and transfection of transient genes.
    Oral presentation and report redaction.
