The proteins of the Calcium Release Complex are precisely localized at the triad, with the RyR and DHPR (2 calcium channels) facing each other in their respective membranes. CRC genes mutations are involved in muscle diseases, and it is now demonstrated that triad mislocalization correlates with muscle contraction defects.
However, the mechanisms responsible for localization and retention of CRC proteins at the triads are still unknown.
The aim of my PhD is to decipher where are CRC proteins synthesized in a muscle cell? How do they traffic to the triads? By which mechanisms are they retained at the triads?
Methods used:
Dissection of skeletal muscle (neonatal mouse)
Primary cultures of mouse skeletal muscle cells (matrigel coating, cellular proliferation and differentiation, freezing...)
Cloning
Lentivirus production and transduction
Cellular transfection (HEK293)
Immunofluorescence
Confocal microscopy (Zeiss LSM 710)
