2008 - 2011Optimization of the piggyBac transposon biosafety for gene transfer: use of mRNA and insulators
My thesis project was to define and validate solutions to improve the biosafety of a non-viral vector, the piggyBac transposon. Gene transfer mediated by piggyBac requires two components: a plasmid carrying the gene of interest to be inserted in the genome, and a source of transposase, the enzyme which catalyzes the transgene integration. One approach of my thesis work was to deliver the source of piggyBac transposase as an mRNA molecule instead of the more commonly-used DNA. This strategy aims to improve the stability of the integration and reduce the genotoxic effects by limiting the presence of the transposase in the cells. The second objective of my project is to improve the expression of the transgene by adding insulator sequences. Indeed, insulators are known to counteract the mechanisms of silencing of the transgene. This strategy reduces the number of integrations required to obtain a sufficient expression of the transgene and thus, improve biosecurity of the system.
