    2013 - 2013 The principal aim of my work was the characterization of the impact on protein-protein interaction of natural non-synonymous Single Nucleotide Polymorphisms (SNPs) present in the human population and/or associated with disease, by "Edgotyping" approach, which entails the systematic determination of the interaction profiles between macromolecules (also called “reference interactome”) followed by the determination of the impact of mutations on known reference interactions (allelic edgotyping). I had to compare the impact of three types of alleles on PPI profiles: natural variants in genes not associated with any disease, natural variants in genes associated with disease, and disease associated mutations. To achieve this goal, a list of genes and their associated variants had to first be generated. The alleles were created by PCR with specifically designed primers, and the PCR products were sequenced to validate that the single nucleotide change had been correctly cloned. The allele bank then inserted into appropriate plasmids for Y2H experiments, and with them an “edgotyping network” could be mapped.

    2012 - 2012 The primary aim of this internship was to perform in vivo testing of transcriptional enhancers in Drosophila embryonic mesoderm. From ongoing studies of enhancers in the lab, mutant enhancers were designed that contain mutations in predicted transcription factor binding sites. I was in charge of examining the effect of these transcription factor binding site mutations on the tissue-specific activity of the tested enhancers in whole Drosophila embryos.
