  • Merck Serono - Senior Technician then Associate Scientist

    Lyon 2001 - maintenant 2001-2004: Senior technician, in charge of automation of biochemical and cellular assays for medium throughput screening. In this role, I developped robotic procedures for various enzymatic assays, using various stand-alone or fully integrated liquid-handling platforms, incubators & readers (CRS robotics, tasks scheduling)
    I contributed to the first in-house validation of robotic cell based assays in the context of a “Functional Genomics” project: apoptosis, survival or proliferation measurements on Jurkat, Neuro2A or Olineu cells, followed by the screening of a library of putative modulator proteins.

    2004-2011: Senior technician then Associate Scientist in the Assay Development & Biochemichal Pharmacology Group. In this role, I was responsible for the development, and validation of a wide range of in-vitro assays for MTS and Compound Profiling.
    I gained direct experience with various assay formats including receptor binding, enzymatic activity, GTPγS assay, as well as expertise with different biological target classes: GPCRs, enzymes (kinases, phosphatases, proteases…).
    I'm now mastering a large panel of detection technologies: direct fluorescence, HTRF, FP, AlphaScreen, radio- isotopes, absorbance, luminescence.
    One of my major accomplishment was the validation & transfer to the Automation Group of an integrated robotized platform for the worldwide profiling of small molecules on P450 Cytochromes
    I'm also responsible for the Caliper Mobility Shift platform, used for development of orthogonal assays to support kinases projects. I evaluated this technology for the determination of compounds mode of action.
    Since June 2011: I was rained in automated electrophysiology (planar patch-clamp), for the study of voltage-gated or ligand-gated ion channels. I performed periodic characterization (biophysics) of new cell lines over-expressing the channels of interest and ensured pharmacology support to several ion channels projects.

    • Determination of the mode of action of Lead candidates
    • Regular support of the SAR for medicinal chemists on several discovery projects, including SOPs writing, data evaluation and posting of the results to the Corporate Database
    • Automation of compounds dilutions & transfer (Caliper Zephyr & Sciclone ALH liquid-handlers) for routine potency determinations or the characterization of more advanced compounds

  • Battelle - Technician then Study Director

    1994 - 2001 I worked for an american contract research organization, in the field of agrochemical products development, first as a technician, then as a Study Director (1998-2001). My functions included the development of analytical methods in the context of pesticides residues studies, various crops samples extraction and analysis with the help of one or two technicians, the redaction of studies protocols and reports, in a GLP environment.
    I performed numerous studies on several crops, using various technologies: assisted samples extraction, solid phase extraction, liquid-liquid partition, HPLC/UV, HPLC/Fluo with post-column derivatization, GC/FID, GC/ECD, LC-MS...
    During my last 3 years in the company, I was identified as the expert responsible for physico-chemical studies, including all studies required for the FDA or OECD registration of new agrochemical products: solubilities, partition coefficient, surface tension, flash point, storage stability, pKa, explodability...
    I gained a lot of experience in implementing official guidelines in the lab, periodic meetings with customers and managing one or two technicians.
    One of my significant contribution to the field was the registration of a new veterinary product for the european market, including the whole physico-chemical package.
  • Battelle - Junior Technician

    1992 - 1994 I conducted several binding studies on numerous receptors, essentially from the central nervous system, for various pharmaceutical companies. For this purpose, I prepared native membranes fractions from rodents brains substructures (or from other organs) and used isotopic labelled ligands. I was autonomous in animals handling, brain dissecting and results reporting. I was also responsible for reagents ordering for my experimental work.
