  • Dr Robert Ballotti - Research Scientist

2014 - maintenant the group of Dr Robert Ballotti. INSERM 1065, C3M, Nice France. The aim of my work was to
determine the role of HACE1 in melanoma development. I assessed melanoma cell viability, migration, adhesion
and proliferation when the target gene was modified (overexpression or invalidation). Using Immunoprecipitation
and Immunoblot techniques, the mechanisms and interactions involved were studied. Through Immunostaining
(IHC and IF), I determined the localization of HACE1 and its co-localization with other proteins using a Leica
microscope. Furthermore, in vivo experiments allowed me to follow the fate of these modified melanomas cells in
"nude" mice using imaging techniques (biospace). Mice lung sections were visualized using a confocal microscope
    (Nikon).
  • Dr Jacques Pouysségur - Student

2010 - 2014 Ph.D student in the group of Dr Jacques Pouysségur under the supervision of Dr Cercina Onesto. UMR 7284,
I explored the cell metabolism reprogramming in polarized macrophages by focusing on the
potential metabolic plasticity of macrophages once in a defined activation state. Using different techniques
(ELISA,Westen Blot, RT-PCR, Immunohistochemistry), I confirmed the phenotype of cells, when and where the
specific macrophages' biomarkers were expressed within the tumor microenvironment, the co-localization of
metabolic biomarkers within the macrophage phenotype. Moreover, I was able to demonstrate some metabolic
plasticity within these polarized macrophages (sea-horse) exist by the targeting of glycolysis through the blockade
of downstream lactic acid export catalyzed by monocarboxylate transporters. By manipulating the metabolic
features of polarized macrophages, we observed that functional polarization of macrophages could be affected by
    changes in cell metabolism.
